reticula\u00e7\u00e3o r\u00e1pida na presen\u00e7a de c\u00e1tions multivalentes (por exemplo, Ca\u00b2\u207a) em solu\u00e7\u00f5es aquosas, resultando em um hidrogel com propriedades mec\u00e2nicas class=\"elementor-heading-title elementor-size-default\">Introdu\u00e7\u00e3o<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-e607aad elementor-widget class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>O alginato \u00e9 um polissacar\u00eddeo presente como componente estrutural das algas e como componente capsular das bact\u00e9rias do solo. Esse polissacar\u00eddeo \u00e9 normalmente obtido a partir de algas marrons e utilizado em diversos setores, como o m\u00e9dico, o farmac\u00eautico, o aliment\u00edcio e o t\u00eaxtil, devido \u00e0s suas propriedades viscosificantes, gelificantes e estabilizantes. O alginato pode sofrer reticula\u00e7\u00e3o rapidamente na presen\u00e7a de c\u00e1tions multivalentes (por exemplo, Ca\u00b2\u207a) em solu\u00e7\u00f5es aquosas, resultando em um hidrogel com propriedades mec\u00e2nicas ajust\u00e1veis. Esse processo simples e econ\u00f4mico, aliado \u00e0 sua capacidade de reter grandes quantidades de l\u00edquido, torna o alginato um biomaterial valioso para aplica\u00e7\u00f5es biotecnol\u00f3gicas, como a administra\u00e7\u00e3o de medicamentos e c\u00e9lulas e a engenharia de tecidos [1].   <\/p><p>Os hidrog\u00e9is de alginato podem ser injetados no corpo ou implantados em um formato pr\u00e9-moldado. No \u00faltimo caso, o alginato tem sido frequentemente processado por meio de impress\u00e3o 3D baseada em extrus\u00e3o, o que requer um processo de gelifica\u00e7\u00e3o r\u00e1pido. O polissacar\u00eddeo oferece gelifica\u00e7\u00e3o r\u00e1pida em condi\u00e7\u00f5es suaves (quando misturado com c\u00e1tions multivalentes) e, dessa forma, as c\u00e9lulas podem ser encapsuladas de maneira f\u00e1cil e r\u00e1pida [2].  <\/p><p>As propriedades viscoel\u00e1sticas do alginato podem ser ajustadas alterando-se diversos par\u00e2metros, tais como a concentra\u00e7\u00e3o e o peso molecular do pol\u00edmero, bem como o tipo e a concentra\u00e7\u00e3o do agente de reticula\u00e7\u00e3o. Nesta nota de aplica\u00e7\u00e3o, o alginato a 1% da CELLINK foi reticulado com solu\u00e7\u00e3o de cloreto de c\u00e1lcio (CaCl\u2082) em diferentes concentra\u00e7\u00f5es (10 mM e 50 mM), e suas propriedades viscoel\u00e1sticas foram medidas no ElastoSens\u2122 Bio ap\u00f3s incuba\u00e7\u00f5es sucessivas com a solu\u00e7\u00e3o. Al\u00e9m disso, a estabilidade mec\u00e2nica de uma formula\u00e7\u00e3o de alginato reticulado (10 mM) foi avaliada ao longo de 24 horas durante a incuba\u00e7\u00e3o com uma solu\u00e7\u00e3o salina (PBS).  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-b169c69 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"b169c69\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Configura\u00e7\u00e3o experimental para avalia\u00e7\u00e3o da reticula\u00e7\u00e3o do hidrogel de alginato\n<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-622c4a9 elementor-widget elementor-widget-heading\" data-id=\"622c4a9\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Materiais e M\u00e9todos<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-37946fb elementor-widget elementor-widget-text-editor\" data-id=\"37946fb\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><a href=\"https:\/\/www.cellink.com\/product\/alginate-5\/\" target=\"_blank\" rel=\"noopener\"><span style=\"font-weight: 400\">Alginato<\/span><\/a><span style=\"font-weight: 400\"> 1% e CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\"> (10 mM e <\/span><a href=\"https:\/\/www.cellink.com\/product\/crosslinking-agent\/\" target=\"_blank\" rel=\"noopener\"><span style=\"font-weight: 400\">50 mM<\/span><\/a><span style=\"font-weight: 400\">) foram adquiridos da CELLINK (Boston, EUA). Foram vertidos 3 mL de alginato no ElastoSens\u2122 <\/span> <span style=\"font-weight: 400\"> O porta-amostra da Bio e 3 mL do <\/span><span style=\"font-weight: 400\">CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\">  A solu\u00e7\u00e3o foi adicionada cuidadosamente sobre a amostra e deixada agir por 15 minutos. Em seguida, a solu\u00e7\u00e3o foi removida para a medi\u00e7\u00e3o das propriedades viscoel\u00e1sticas do alginato no ElastoSens\u2122 <\/span><span style=\"font-weight: 400\">  Bio a 25 \u00b0C por 5 minutos (com um intervalo de 10 segundos). Esse \u00faltimo procedimento foi repetido durante as primeiras duas horas. Para as amostras reticuladas com    <\/span><span style=\"font-weight: 400\">CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\">  A 10 mM, a solu\u00e7\u00e3o foi deixada durante a noite \u00e0 temperatura ambiente e uma \u00faltima medi\u00e7\u00e3o foi realizada aproximadamente 24 horas ap\u00f3s a remo\u00e7\u00e3o da solu\u00e7\u00e3o. Em seguida, adicionou-se PBS sem c\u00e1lcio a essas amostras para avaliar sua dissolu\u00e7\u00e3o ao longo do tempo a 37 \u00b0C. As medi\u00e7\u00f5es foram realizadas a cada hora durante as primeiras 6 horas (durante 5 minutos, com intervalos de 10 segundos) e uma \u00faltima medi\u00e7\u00e3o foi realizada ap\u00f3s 24 horas de incuba\u00e7\u00e3o. Os resultados m\u00e9dios s\u00e3o expressos como m\u00e9dia \u00b1 desvio-padr\u00e3o. O n\u00famero de amostras foi igual a 3 (n = 3).      <\/span><span style=\"font-weight: 400\">As an\u00e1lises estat\u00edsticas foram realizadas utilizando o GraphPad Prism vers\u00e3o 9 (software GraphPad Prism, La Jolla, CA, EUA). As compara\u00e7\u00f5es entre os grupos foram avaliadas por ANOVA unidirecional com teste post hoc de Tukey para corre\u00e7\u00e3o de compara\u00e7\u00f5es m\u00faltiplas. Considerou-se que havia signific\u00e2ncia quando p &lt; e a 0,05.  <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-c74affc elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"c74affc\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Efeitos da concentra\u00e7\u00e3o de cloreto de c\u00e1lcio nas propriedades do hidrogel de alginato\n<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-2ba4198 elementor-widget elementor-widget-heading\" data-id=\"2ba4198\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Resultados e Discuss\u00e3o<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-16b71b6 elementor-widget elementor-widget-text-editor\" data-id=\"16b71b6\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Tr\u00eas amostras de cada condi\u00e7\u00e3o de reticula\u00e7\u00e3o (10 mM e 50 mM de CaCl\u2082) foram testadas periodicamente no ElastoSens\u2122 Bio. Os valores m\u00e9dios em cada ponto temporal do m\u00f3dulo de armazenamento de cisalhamento G\u2019 (Pa) e da altura da amostra (mm) do alginato com solu\u00e7\u00e3o de 10 mM de CaCl\u2082 est\u00e3o representados nas Figuras 1 e 2. Para as amostras reticuladas com CaCl\u2082 a 10 mM, os g\u00e9is incharam nos momentos iniciais, o que \u00e9 confirmado pelo aumento em sua altura m\u00e9dia. A altura apresentou um aumento menos pronunciado ap\u00f3s 45 minutos a 4 horas de incuba\u00e7\u00e3o (Fig. 3A). A altura final ap\u00f3s 24 horas mostrou que as amostras ainda apresentavam um incha\u00e7o significativo (68% maior do que a altura inicial). A ligeira diminui\u00e7\u00e3o do G\u2019 nos primeiros 30 minutos sugere que o incha\u00e7o foi mais significativo do que as liga\u00e7\u00f5es cruzadas formadas at\u00e9 aquele momento. Depois disso, o G\u2019 apresentou um aumento suave e esperado com o tempo, atingindo 645 \u00b1 55 Pa ap\u00f3s 4 horas de incuba\u00e7\u00e3o com a solu\u00e7\u00e3o de CaCl\u2082 (com diferen\u00e7as estat\u00edsticas a cada 15 minutos). O valor final de G\u2019 dos g\u00e9is reticulados ap\u00f3s 24 horas de incuba\u00e7\u00e3o foi igual a 887 \u00b1 9 Pa. O Tan(\u03b4) mostrou-se est\u00e1vel de 15 minutos at\u00e9 4 horas de incuba\u00e7\u00e3o (~0,15); no entanto, apresentou um aumento ap\u00f3s 24 horas, atingindo o valor de 0,3 (dados n\u00e3o mostrados). Esse aumento sugere que a fra\u00e7\u00e3o viscosa do hidrogel tornou-se mais predominante com o tempo.         <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element Devido a esse comportamento, o teste foi interrompido assim que a amostra perdeu completamente o contato com a parede interna do suporte de amostras (Fig. 3B), o que \u00e9 um requisito do teste. O valor de G\u2019 aumentou com o tempo, atingindo o n\u00edvel de 2.832 \u00b1 53 Pa ap\u00f3s 1,5 horas de incuba\u00e7\u00e3o (com diferen\u00e7as estat\u00edsticas a cada 5 minutos). A dissolu\u00e7\u00e3o do alginato por meio da libera\u00e7\u00e3o de \u00edons c\u00e1lcio no PBS foi significativa durante as primeiras 24 horas de incuba\u00e7\u00e3o a 37 \u00b0C, conforme demonstrado pela diminui\u00e7\u00e3o de G\u2019 com o tempo (redu\u00e7\u00e3o de 20% ap\u00f3s 24 horas).
