de quantifica\u00e7\u00e3o celular vai al\u00e9m das c\u00e2maras de contagem tradicionais, levando ao desenvolvimento de solu\u00e7\u00f5es inovadoras que facilitam a contagem precisa e, ao mesmo tempo, reduzem o tempo necess\u00e1rio.  <\/li><li>A turbidez, que reflete a opacidade da solu\u00e7\u00e3o na presen\u00e7a de part\u00edculas que dispersam a luz, surge como um meio eficaz para acelerar a quantifica\u00e7\u00e3o celular.<\/li><li>Nesta nota de aplica\u00e7\u00e3o, foi estabelecida uma forte correla\u00e7\u00e3o entre a turbidez (FTU) de uma solu\u00e7\u00e3o de cultura <em>de S. cerevisiae<\/em> e o n\u00famero de c\u00e9lulas (c\u00e9lulas\/mL) contadas com uma c\u00e2mara de contagem de c\u00e9lulas convencional.  <\/li><li>O uso <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">do TURBIDI.T\u2122<\/a> para medi\u00e7\u00f5es precisas de turbidez oferece uma abordagem eficientes de quantifica\u00e7\u00e3o celular levou \u00e0 busca por alternativas \u00e0 t\u00e9cnica tradicional e demorada das c\u00e2maras de contagem de c\u00e9lulas. Essa abordagem convencional, embora confi\u00e1vel, apresenta desafios em termos de velocidade e praticidade, especialmente diante das crescentes exig\u00eancias da pesquisa e da ind\u00fastria. <\/p><p>A turbidez, uma medida da opacidade de uma solu\u00e7\u00e3o, surge como uma abordagem interessante para acelerar a quantifica\u00e7\u00e3o celular. O n\u00edvel de turbidez em um meio tem sido associado \u00e0 concentra\u00e7\u00e3o celular em uma cultura celular [1]. Aproveitando essa rela\u00e7\u00e3o, esta nota de aplica\u00e7\u00e3o apresenta uma nova abordagem para determinar rapidamente o n\u00famero de c\u00e9lulas. Esse m\u00e9todo utiliza os recursos do <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122<\/a>, um instrumento compat\u00edvel com a IoT projetado para a medi\u00e7\u00e3o precisa da turbidez. Foi inoculado um volume de 100 mL, a fim de obter um valor inicial de turbidez de 200 FTU. Ap\u00f3s a inocula\u00e7\u00e3o, foram coletadas al\u00edquotas est\u00e9reis a cada duas horas e o n\u00famero de c\u00e9lulas (ou seja, a concentra\u00e7\u00e3o celular) foi determinado pelo m\u00e9todo de colora\u00e7\u00e3o com azul de metileno. Esse m\u00e9todo se baseia em um corante, o azul de metileno, que se liga aos componentes celulares e os tinge de azul. Esse corante reage de maneira diferente em c\u00e9lulas vi\u00e1veis e n\u00e3o vi\u00e1veis, pois \u00e9 transformado pelas c\u00e9lulas vivas (que permanecem incolores), enquanto as c\u00e9lulas mortas permanecem coradas de azul. As c\u00e9lulas coradas foram colocadas em um <a href=\"https:\/\/en.wikipedia.org\/wiki\/Hemocytometer\" target=\"_blank\" rel=\"noopener\">hemocit\u00f4metro<\/a> (tamb\u00e9m conhecido como c\u00e2mara de contagem de c\u00e9lulas ou c\u00e2mara de Malassez) e observadas ao microsc\u00f3pio (x40).      O hemocit\u00f4metro \u00e9 composto por nove quadrados de 1 x 1 mm (1 mm\u00b2). Utilizou-se um volume de 1 \u00b5L para cada medi\u00e7\u00e3o.   <\/p><p>A turbidez das solu\u00e7\u00f5es foi determinada com o uso do <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122.<\/a> O instrumento foi calibrado com padr\u00f5es de formazina (1\u20134.000 FTU). A emiss\u00e3o e a recep\u00e7\u00e3o de luz foram realizadas utilizando <a href=\"https:\/\/rheolution-store.com\/collections\/cartridges\/products\/emitt-850\" target=\"_blank\" rel=\"noopener\">cartuchos Emitt.805<\/a> (850 nm) e <a href=\"https:\/\/rheolution-store.com\/collections\/cartridges\/products\/cartridge-receiv-vis\" target=\"_blank\" rel=\"noopener\">cartuchos Receiv.ViS<\/a> (comprimento de onda variando de 400 nm a 1.000 nm), respectivamente.   Utilizou-se um comprimento de onda de 850 nm, uma vez que o efeito da absor\u00e7\u00e3o de luz pela amostra era menor (dados n\u00e3o apresentados). Neste trabalho, optamos por utilizar frascos de 10 mL, nos quais as amostras foram al\u00edquotadas e medidas no <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122.<\/a>  Os dados foram coletados e exibidos no tablet no qual estava instalado o aplicativo Soft Matter Analytics App\u2122.<\/p><p>Tanto a contagem de c\u00e9lulas quanto as medi\u00e7\u00f5es de turbidez foram realizadas em triplicata (n = 3). Os dados s\u00e3o apresentados como m\u00e9dia \u00b1 desvio-padr\u00e3o. A variabilidade \u00e9 definida como a raz\u00e3o entre o desvio-padr\u00e3o e o valor m\u00e9dio.  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-270ac45 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"270ac45\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">AN\u00c1LISE COMPARATIVA E CORRELA\u00c7\u00c3O ENTRE OS INDICADORES DE TURBIDIDADE E DE CONTAGEM DE C\u00c9LULAS<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-88497d3 elementor-widget elementor-widget-heading\" data-id=\"88497d3\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Resultados e Discuss\u00e3o<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-a479882 elementor-widget elementor-widget-text-editor\" data-id=\"a479882\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Al\u00edquotas celulares na fase de crescimento da cultura de levedura foram analisadas por meio da medi\u00e7\u00e3o da turbidez e da contagem de c\u00e9lulas com uma c\u00e2mara de contagem. <b>A Figura 3<\/b> mostra a evolu\u00e7\u00e3o temporal de ambos os par\u00e2metros, que aumentam com o tempo. Observamos no gr\u00e1fico que a evolu\u00e7\u00e3o da turbidez e da concentra\u00e7\u00e3o celular segue a mesma tend\u00eancia, que \u00e9, em geral, exponencial. O aumento da concentra\u00e7\u00e3o celular est\u00e1 afetando diretamente a turbidez da cultura. Mais c\u00e9lulas est\u00e3o espalhando a luz e afetando a turbidez geral do meio.   <\/p><p>As medi\u00e7\u00f5es de turbidez foram mais consistentes (apresentando 3,8% de variabilidade) em compara\u00e7\u00e3o com a t\u00e9cnica de contagem de c\u00e9lulas (com uma variabilidade de 17,6%). Essa diverg\u00eancia pode ser atribu\u00edda a imprecis\u00f5es na contagem decorrentes da percep\u00e7\u00e3o humana, mas tamb\u00e9m \u00e0 diferen\u00e7a no volume da amostra entre os dois m\u00e9todos. Enquanto a contagem de c\u00e9lulas utiliza 1 \u00b5L de amostra, a avalia\u00e7\u00e3o da turbidez \u00e9 realizada neste estudo com al\u00edquotas de 10 mL. O gr\u00e1fico obtido pode ent\u00e3o ser utilizado para estimar a contagem de c\u00e9lulas em uma amostra, bastando para isso correlacionar o valor de turbidez medido com <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">o TURBIDI.T\u2122<\/a> ao n\u00famero de c\u00e9lulas associado. an\u00e1lise da contagem de c\u00e9lulas realizada com uma c\u00e2mara de contagem convencional apresenta alta correla\u00e7\u00e3o com os valores de turbidez (R\u00b2 = 0,998), demonstrando que \u00e9 poss\u00edvel desenvolver e utilizar com efic\u00e1cia um m\u00e9todo de infer\u00eancia para estimar o n\u00famero de c\u00e9lulas em um meio de cultura.<\/li><li>Os recursos de
