EXPLORANDO SEU PAPEL NA ESPECTROFOTOMETRIA <\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-7144aa8 elementor-widget elementor-widget-text-editor\" data-id=\"7144aa8\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>A absorv\u00e2ncia \u00e9 um conceito fundamental no campo da espectrofotometria, t\u00e9cnica utilizada para medir a quantidade de luz absorvida por uma subst\u00e2ncia em uma ampla faixa de comprimentos de onda. Trata-se de uma grandeza adimensional derivada do logaritmo da raz\u00e3o entre a luz incidente e a luz transmitida atrav\u00e9s de uma amostra. O espectro de absorv\u00e2ncia, frequentemente representado por um gr\u00e1fico, revela os comprimentos de onda nos quais uma subst\u00e2ncia absorve a luz com maior intensidade (<strong>Figura 1<\/strong>). Os valores de absorv\u00e2ncia em um determinado comprimento de onda s\u00e3o amplamente utilizados em an\u00e1lises qu\u00edmicas e bioqu\u00edmicas para quantificar a concentra\u00e7\u00e3o de uma subst\u00e2ncia em solu\u00e7\u00e3o ou para monitorar rea\u00e7\u00f5es ao longo do tempo. Os diferentes cartuchos de emiss\u00e3o (como Emitt.405, Emitt.520, Emitt.635, Emitt.850) permitem que o usu\u00e1rio selecione o comprimento de onda adequado para a subst\u00e2ncia em quest\u00e3o. Seu pico de absor\u00e7\u00e3o ocorre a 668 e 609 nm (Merck Index). Diferentes concentra\u00e7\u00f5es de azul de metileno em \u00e1gua desionizada foram testadas no <a href=\"https:\/\/rheolution.com\/products\/absor-b\/\" target=\"_blank\" rel=\"noopener\">ABSOR.B\u2122<\/a> utilizando frascos de vidro transparente de 1 mL e os cartuchos Emitt.405, Emitt.520, Emitt.635 e Emitt.850.    <\/p><p><strong>A Figura 2<\/strong> mostra os resultados m\u00e9dios \u00b1 desvio padr\u00e3o (tr\u00eas amostras diferentes por concentra\u00e7\u00e3o testada, n=3). Conforme esperado a partir do espectro (setas na Figura 1), a luz vermelha (635 nm) foi fortemente absorvida, seguida por uma leve absor\u00e7\u00e3o da luz verde (520 nm). As luzes violeta-azul (405 nm) e infravermelha (850 nm) apresentaram absor\u00e7\u00e3o quase nula na faixa de concentra\u00e7\u00f5es estudada.  <\/p><p>A rela\u00e7\u00e3o entre a absorv\u00e2ncia e a concentra\u00e7\u00e3o \u00e9 linear, de acordo com a <a href=\"https:\/\/chem.libretexts.org\/Bookshelves\/Physical_and_Theoretical_Chemistry_Textbook_Maps\/Supplemental_Modules_(Physical_and_Theoretical_Chemistry)\/Spectroscopy\/Electronic_Spectroscopy\/Electronic_Spectroscopy_Basics\/The_Beer-Lambert_Law\" target=\"_blank\" rel=\"noopener\">Lei de Beer-Lambert<\/a>, desde que a solu\u00e7\u00e3o n\u00e3o seja muito concentrada e que a luz incidente seja monocrom\u00e1tica. A rela\u00e7\u00e3o linear obtida com a luz vermelha pode ser utilizada para determinar a concentra\u00e7\u00e3o de azul de metileno em uma solu\u00e7\u00e3o desconhecida. <\/p><p>De acordo com os resultados de absorv\u00e2ncia, a transmit\u00e2ncia da luz vermelha (635 nm) diminuiu \u00e0 medida que a concentra\u00e7\u00e3o de azul de metileno na solu\u00e7\u00e3o aumentou. A 10 mg\/L, menos de 10% da luz era transmitida. A transmit\u00e2ncia apresentou uma ligeira diminui\u00e7\u00e3o para as luzes verde (520 nm), azul-violeta (405 nm) e infravermelha (850 nm) (sempre superior a 85%).
