ricavato dalle alghe, ampiamente utilizzato per il rilascio di cellule e proteine e per applicazioni nell&#8217;ingegneria tissutale.<\/li><li>L&#8217;alginato si reticola rapidamente in presenza di cationi multivalenti (ad es. Questo polisaccaride viene solitamente ricavato dalle alghe brune e viene utilizzato in molti settori, come quello medico, farmaceutico, alimentare e tessile, grazie alle sue propriet\u00e0 viscosizzanti, gelificanti e stabilizzanti. L\u2019alginato pu\u00f2 reticolarsi rapidamente in presenza di cationi multivalenti (ad es. Ca\u00b2\u207a) in soluzioni acquose, dando origine a un idrogel con propriet\u00e0 meccaniche regolabili. Questo processo semplice ed economico, unito alla sua capacit\u00e0 di trattenere grandi quantit\u00e0 di liquido, rende l\u2019alginato un biomateriale prezioso per applicazioni biotecnologiche come il rilascio di farmaci e cellule e l\u2019ingegneria tissutale [1].   <\/p><p>Gli idrogel di alginato possono essere iniettati nel corpo o impiantati in una forma predefinita. In quest\u2019ultimo caso, l\u2019alginato viene spesso lavorato tramite stampa 3D a estrusione, che richiede un processo di gelificazione veloce. Questo polisaccaride gelifica rapidamente in condizioni moderate (quando viene miscelato con cationi multivalenti) e, in questo modo, le cellule possono essere incapsulate facilmente e velocemente [2].  <\/p><p>Le propriet\u00e0 viscoelastiche dell\u2019alginato possono essere regolate modificando diversi parametri, come la concentrazione e il peso molecolare del polimero, nonch\u00e9 il tipo e la concentrazione dell\u2019agente reticolante. In questa nota applicativa, l\u2019alginato all\u20191% di CELLINK \u00e8 stato reticolato con una soluzione di cloruro di calcio (CaCl\u2082) a diverse concentrazioni (10 mM e 50 mM) e le sue propriet\u00e0 viscoelastiche sono state misurate con ElastoSens\u2122 Bio dopo incubazioni successive con la soluzione. Inoltre, \u00e8 stata valutata la stabilit\u00e0 meccanica di una formulazione di alginato reticolato (10 mM) nell\u2019arco di 24 ore durante l\u2019incubazione con una soluzione salina (PBS).  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-b169c69 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"b169c69\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Configurazione sperimentale per la valutazione della reticolazione dell'idrogel di alginato\n<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-622c4a9 elementor-widget elementor-widget-heading\" data-id=\"622c4a9\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Materiali e metodi<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-37946fb elementor-widget elementor-widget-text-editor\" data-id=\"37946fb\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><a href=\"https:\/\/www.cellink.com\/product\/alginate-5\/\" target=\"_blank\" rel=\"noopener\"><span style=\"font-weight: 400\">Alginato<\/span><\/a><span style=\"font-weight: 400\"> 1% e CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\"> (10 mM e <\/span><a href=\"https:\/\/www.cellink.com\/product\/crosslinking-agent\/\" target=\"_blank\" rel=\"noopener\"><span style=\"font-weight: 400\">50 mM<\/span><\/a><span style=\"font-weight: 400\">) sono stati acquistati da CELLINK (Boston, USA). Sono stati versati 3 mL di alginato nell&#8217;ElastoSens\u2122 <\/span> <span style=\"font-weight: 400\"> Il portacampioni Bio e 3 mL del <\/span><span style=\"font-weight: 400\">CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\">  La soluzione \u00e8 stata versata delicatamente sulla superficie del campione e lasciata agire per 15 minuti. La soluzione \u00e8 stata poi rimossa per misurare le propriet\u00e0 viscoelastiche dell\u2019alginato con ElastoSens\u2122 <\/span><span style=\"font-weight: 400\">  Bio a 25 \u00b0C per 5 minuti (con un intervallo temporale di 10 secondi). Quest\u2019ultima procedura \u00e8 stata ripetuta per le prime due ore. Per i campioni reticolati con    <\/span><span style=\"font-weight: 400\">CaCl<\/span><span style=\"font-weight: 400\">2<\/span><span style=\"font-weight: 400\">  A 10 mM, la soluzione \u00e8 stata lasciata riposare per tutta la notte a temperatura ambiente e un\u2019ultima misurazione \u00e8 stata effettuata circa 24 ore dopo aver rimosso la soluzione. A questi campioni \u00e8 stato poi aggiunto PBS privo di calcio per valutare la loro dissoluzione nel tempo a 37 \u00b0C. Le misurazioni sono state effettuate ogni ora per le prime 6 ore (per 5 minuti con un intervallo temporale di 10 secondi) e un\u2019ultima misurazione \u00e8 stata effettuata dopo 24 ore di incubazione. I risultati medi sono espressi come media \u00b1 deviazione standard. Il numero di campioni era pari a 3 (n = 3).      <\/span><span style=\"font-weight: 400\">Le analisi statistiche sono state effettuate utilizzando GraphPad Prism versione 9 (GraphPad Prism software, La Jolla, CA, USA). I confronti tra i gruppi sono stati valutati mediante ANOVA a una via con test post-hoc di Tukey per correggere i confronti multipli. Si \u00e8 considerato significativo un valore di p &lt; e a 0,05.  <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-c74affc elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"c74affc\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Effetti della concentrazione di cloruro di calcio sulle propriet\u00e0 dell'idrogel di alginato\n<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-2ba4198 elementor-widget elementor-widget-heading\" data-id=\"2ba4198\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Risultati e discussione<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-16b71b6 elementor-widget elementor-widget-text-editor\" data-id=\"16b71b6\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Sono stati analizzati periodicamente nell\u2019ElastoSens\u2122 Bio tre campioni per ciascuna condizione di reticolazione (10 mM e 50 mM di CaCl\u2082). I valori medi in ciascun intervallo di tempo del modulo di immagazzinamento al taglio G\u2019 (Pa) e dell\u2019altezza del campione (mm) dell\u2019alginato con soluzione di CaCl\u2082 a 10 mM sono riportati nelle figure 1 e 2. Per i campioni reticolati con 10 mM di CaCl\u2082, i gel si sono gonfiati nei primi momenti, come conferma l\u2019aumento della loro altezza media. L\u2019altezza ha mostrato un aumento meno marcato dopo 45 minuti e fino a 4 ore di incubazione (Fig. 3A). L\u2019altezza finale dopo 24 ore ha mostrato che i campioni presentavano ancora un rigonfiamento significativo (superiore del 68% rispetto all\u2019altezza iniziale). La leggera diminuzione del G\u2019 nei primi 30 minuti suggerisce che il rigonfiamento fosse pi\u00f9 significativo dei legami di reticolazione formatisi fino a quel momento. Successivamente, il valore di G\u2019 ha mostrato un aumento graduale e previsto nel tempo, raggiungendo 645 \u00b1 55 Pa dopo 4 ore di incubazione con la soluzione di CaCl\u2082 (con differenze statistiche ogni 15 minuti). Il valore finale di G\u2019 dei gel reticolati dopo 24 ore di incubazione era pari a 887 \u00b1 9 Pa. Il Tan(\u03b4) si \u00e8 dimostrato stabile da 15 minuti fino a 4 ore di incubazione (~ 0,15), ma ha mostrato un aumento dopo 24 ore, raggiungendo il valore di 0,3 (dati non mostrati). Questo aumento suggerisce che la componente viscosa dell\u2019idrogel sia diventata pi\u00f9 predominante con il passare del tempo.         <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div A causa di questo comportamento, il test \u00e8 stato interrotto non appena il campione ha perso completamente il contatto con la parete interna del portacampioni (Fig. 3B), come previsto dal protocollo. Il valore di G\u2019 \u00e8 aumentato col passare del tempo, raggiungendo il livello di 2832 \u00b1 53 Pa dopo 1,5 ore di incubazione (con differenze statistiche ogni 5 minuti). L\u2019altezza \u00e8 diminuita col passare del tempo (a causa del restringimento) e il valore di tan(\u03b4) \u00e8 rimasto stabile (~ 0,15). Questo era legato alla velocit\u00e0 di reticolazione, che aumentava all\u2019aumentare della concentrazione di calcio.
