Questa struttura unica, che unisce elementi inorganici e organici, conferisce al PDMS un\u2019eccezionale mobilit\u00e0 della catena, una temperatura di transizione vetrosa molto bassa e un comportamento elastomerico stabile in un ampio intervallo di temperature. Il PDMS viene prodotto esclusivamente tramite sintesi industriale, che in genere prevede l\u2019idrolisi e la condensazione di clorosilani, seguite dalla polimerizzazione ad apertura di anello dei silossani ciclici.  <\/span><\/p><p><span style=\"font-weight: 400\">Il PDMS \u00e8 intrinsecamente <\/span><b>idrofobico<\/b><span style=\"font-weight: 400\">  e non assorbe acqua n\u00e9 si gonfia. Di conseguenza,   <\/span><b>Il PDMS di per s\u00e9 non \u00e8 un idrogel<\/b><span style=\"font-weight: 400\">, bens\u00ec un <\/span><b>elastomero siliconico<\/b><span style=\"font-weight: 400\">. Tuttavia, il PDMS viene spesso incorporato in  <\/span><b>sistemi polimerici ibridi<\/b><span style=\"font-weight: 400\">, come le reti polimeriche interpenetranti o gli idrogel siliconici, in cui una fase polimerica idrofila permette l\u2019assorbimento d\u2019acqua mentre il PDMS garantisce elasticit\u00e0, permeabilit\u00e0 e stabilit\u00e0 meccanica a lungo termine.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-5703c6a elementor-widget__width-initial elementor-widget-mobile__width-inherit del legame silossanico.<\/span><\/p><ul><li style=\"font-weight: 400\"><b>Meccanismi di polimerizzazione:<\/b><span style=\"font-weight: 400\"> Le catene di PDMS si formano attraverso la polimerizzazione con apertura dell&#8217;anello dei silossani ciclici.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><b>Strategie di reticolazione:<\/b><b><br><\/b><ul><li style=\"font-weight: 400\"><span style=\"font-weight: 400\">Indurimento per condensazione tramite reazioni silanoliche.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><span style=\"font-weight: 400\">Polimerizzazione per addizione tramite idrosililazione tra silossani con gruppi funzionali vinilici e idrurici.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><span style=\"font-weight: 400\">Reticolazione radicale o fotoindotta, comprese le reazioni tiolo-ene.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><\/ul><\/li><li style=\"font-weight: 400\"><b>Formazione di materiali ibridi:<\/b><span style=\"font-weight: 400\"> Il PDMS pu\u00f2 essere combinato con polimeri idrofili per formare idrogel di silicone o reti polimeriche interpenetranti in grado di assorbire acqua.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><\/ul><p><span style=\"font-weight: 400\">La densit\u00e0 di reticolazione, la scelta del catalizzatore e l&#8217;architettura della rete influenzano fortemente l&#8217;elasticit\u00e0, la permeabilit\u00e0 e il comportamento viscoelastico.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-30981bc meccaniche morbide, simili alla gomma, caratterizzate da risposte elastiche e viscoelastiche.<\/span><\/p><ul><li style=\"font-weight: 400\"><b>Elasticit\u00e0:<\/b><span style=\"font-weight: 400\"> La struttura flessibile a base di silossano permette grandi deformazioni reversibili con un&#8217;isteresi minima.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><b>Rigidit\u00e0 regolabile:<\/b><span style=\"font-weight: 400\"> Le propriet\u00e0 meccaniche possono essere regolate in un ampio intervallo modificando la densit\u00e0 dei legami incrociati, il peso molecolare tra i legami incrociati o incorporando reti polimeriche secondarie.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><b>Comportamento viscoelastico:<\/b><span style=\"font-weight: 400\"> Il PDMS mostra un rilassamento delle sollecitazioni dipendente dal tempo e una dissipazione di energia sotto carico ciclico o prolungato.<\/span><\/li><li style=\"font-weight: 400\"><b>Stabilit\u00e0 a lungo termine:<\/b> la struttura a base di silossano, che non si degrada, garantisce un\u2019eccellente resistenza meccanica rispetto a molti sistemi a base di idrogel.<\/li><\/ul>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-0959975 elementor-widget delle proteine e l&#8217;adesione delle cellule; per favorire interazioni cellulari controllate, si ricorre comunemente alla modifica della superficie o all&#8217;ibridazione con polimeri idrofili.<\/span><span style=\"font-weight: 400\"><br><\/span><\/li><li style=\"font-weight: 400\"><b>Immunogenicit\u00e0:<\/b><span style=\"font-weight: 400\"> Il PDMS provoca in genere una risposta infiammatoria minima nelle applicazioni biomediche.<\/span><\/li><li style=\"font-weight: 400\"><b>Degradazione:<\/b><span> Il PDMS non \u00e8 degradabile enzimaticamente, il che ne favorisce l&#8217;uso a lungo termine ma ne limita le applicazioni che richiedono il ricambio della matrice.<\/span><\/li><\/ul>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element style=\"font-weight: 400\">Il PDMS \u00e8 ampiamente utilizzato come <\/span><b>substrato elastico, componente di scaffold e materiale strutturale<\/b><span style=\"font-weight: 400\">  nell&#8217;ingegneria dei tessuti. La sua rigidit\u00e0 regolabile, la trasparenza ottica e la compatibilit\u00e0 con le tecniche di micro e nanofabbricazione permettono un controllo preciso degli stimoli meccanici e della topografia superficiale. In molti sistemi, il PDMS funge da supporto meccanicamente stabile per gli idrogel che offrono un ambiente interattivo per le cellule.  <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-d01f123 elementor-widget elementor-widget-heading\" data-id=\"d01f123\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h3 class=\"elementor-heading-title elementor-size-default\">Coltura cellulare 3D e modelli di malattia<\/h3>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-137d5ec elementor-widget elementor-widget-text-editor\" data-id=\"137d5ec\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"font-weight: 400\">Il PDMS \u00e8 un materiale fondamentale per <\/span><b>dispositivi microfluidici e piattaforme &#8220;organ-on-chip&#8221;<\/b><span style=\"font-weight: 400\">. Anche se il PDMS di per s\u00e9 non \u00e8 un idrogel, viene spesso combinato con compartimenti in idrogel per creare sistemi di coltura 3D in cui \u00e8 possibile controllare con precisione il trasporto dei nutrienti, il confinamento meccanico e i gradienti biochimici.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-c1cb3eb elementor-widget elementor-widget-heading\" data-id=\"c1cb3eb\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h3 class=\"elementor-heading-title elementor-size-default\">Somministrazione di farmaci, geni e cellule<\/h3>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-ff74672 elementor-widget elementor-widget-text-editor\" data-id=\"ff74672\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"font-weight: 400\">PDMS fa parte dei <\/span><b>sistemi a base di silicone idrogel e reti polimeriche interpenetranti<\/b><span style=\"font-weight: 400\">  utilizzati per applicazioni di somministrazione controllata. Il rilassamento delle sollecitazioni e lo smorzamento in funzione del tempo influenzano il modo in cui le forze vengono trasmesse e dissipate sotto carico continuo o ciclico. Nelle applicazioni biomediche e di meccanobiologia, queste propriet\u00e0 viscoelastiche influenzano la meccanosensibilit\u00e0 cellulare, la stabilit\u00e0 meccanica a lungo termine e la resistenza alla fatica. Nei sistemi a idrogel contenenti PDMS, la risposta viscoelastica della fase PDMS gioca un ruolo fondamentale nel definire l\u2019ambiente meccanico composito in cui si trovano le cellule.   <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-851f298 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"851f298\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Metodi per caratterizzare la viscoelasticit\u00e0 dei polimeri a base di PDMS<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-171be53 elementor-widget elementor-widget-text-editor\" data-id=\"171be53\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"font-weight: 400\">Le propriet\u00e0 meccaniche e viscoelastiche del PDMS vengono solitamente caratterizzate tramite prove di trazione, prove di compressione, reometria e analisi meccanica dinamica. Queste tecniche permettono di misurare il modulo elastico, i moduli di immagazzinamento e di perdita, e il comportamento di smorzamento. Monitorando le variazioni nella risposta vibrazionale, permette di misurare con precisione le propriet\u00e0 meccaniche senza deformare fisicamente il campione. Questo approccio \u00e8 particolarmente adatto ai polimeri morbidi a base di PDMS e ai sistemi di idrogel contenenti PDMS.  <\/span><\/p><p><span style=\"font-weight: 400\">Tra i principali vantaggi ci sono i test non distruttivi, il monitoraggio in tempo reale, l&#8217;elevata sensibilit\u00e0 e ripetibilit\u00e0, e la compatibilit\u00e0 con flussi di lavoro sterili o contenenti cellule. La sezione seguente illustra i materiali e i metodi utilizzati, seguiti dai risultati, fornendo un esempio pratico della capacit\u00e0 dello strumento di monitorare in modo non distruttivo le propriet\u00e0 viscoelastiche nel tempo. <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-ed23fc9 elementor-widget elementor-widget-heading\" data-id=\"ed23fc9\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h3 class=\"elementor-heading-title elementor-size-default\">Materiali e metodi<\/h3>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-d118051 elementor-widget elementor-widget-text-editor\" data-id=\"d118051\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"font-weight: 400\">Il polidimetilsilossano (PDMS) \u00e8 stato preparato utilizzando Sylgard 527, composto da una base di PDMS con terminazioni viniliche (Parte A) e da un agente indurente contenente idrosilano con catalizzatore al platino (Parte B). I due componenti sono stati miscelati in un rapporto ponderale di 1:1. Prima della prova non \u00e8 stata eseguita alcuna fase di degassamento.    <\/span><\/p><p><span style=\"font-weight: 400\">Subito dopo la miscelazione, il PDMS non indurito \u00e8 stato versato direttamente nel portacampioni macro (volume totale di 2,0 mL). Per evitare che la superficie si seccasse e che il liquido evaporasse durante i test a lungo termine, la superficie del campione \u00e8 stata ricoperta con un liquido antievaporazione. <\/span><\/p><p><span style=\"font-weight: 400\">La cinetica di indurimento e l&#8217;evoluzione della rigidit\u00e0 complessiva sono state monitorate per un periodo di 24 ore a 70 \u00b0C con il sistema ElastoSens Bio. Le misurazioni sono state effettuate ogni 20 secondi, consentendo di rilevare sia il comportamento di indurimento nelle fasi iniziali sia l&#8217;avvicinamento alla rigidit\u00e0 finale di plateau. La configurazione sperimentale \u00e8 illustrata nella Figura 1.  <\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t<div class=\"elementor-element elementor-element-4133609 e-grid e-con-full campioni di PDMS Sylgard 527 in rapporto 1:1 \u00e8 illustrata nella Figura 2. Durante tutto il periodo di polimerizzazione di 24 ore, sia il modulo di immagazzinamento al taglio (G\u2032) che il modulo di perdita al taglio (G\u2033) sono aumentati, in linea con la progressiva reticolazione e la formazione della rete. Alla fine dell\u2019esperimento, G\u2032 ha raggiunto 7048 \u00b1 43 Pa, mentre G\u2033 si \u00e8 stabilizzato a 2420 \u00b1 167 Pa. Il valore finale pi\u00f9 elevato di G\u2032 rispetto a G\u2033 indica che la risposta del materiale indurito era prevalentemente elastica, cio\u00e8 l\u2019accumulo di energia nella rete reticolata ha prevalso sulla dissipazione viscosa di energia.   <\/span><\/p><p><span style=\"font-weight: 400\">Il fattore di smorzamento (tan \u03b4 = G\u2033\/G\u2032) ha raggiunto in media il valore massimo a 9,54 min.
