Il s&rsquo;agit d&rsquo;une grandeur sans dimension d\u00e9riv\u00e9e du logarithme du rapport entre la lumi\u00e8re incidente et la lumi\u00e8re transmise \u00e0 travers un \u00e9chantillon. Le spectre d\u2019absorbance, souvent repr\u00e9sent\u00e9 sous forme de graphique, r\u00e9v\u00e8le les longueurs d\u2019onde auxquelles une substance absorbe le plus fortement la lumi\u00e8re (<strong>figure 1<\/strong>). Cet instrument moderne mesure la lumi\u00e8re absorb\u00e9e par une substance \u00e0 des longueurs d&rsquo;onde discr\u00e8tes \u00e0 l&rsquo;aide de cartouches optiques interchangeables \u00e9mettant une lumi\u00e8re monochromatique. Les valeurs d\u2019absorbance \u00e0 une longueur d\u2019onde donn\u00e9e sont largement utilis\u00e9es en analyse chimique et biochimique pour quantifier la concentration d\u2019une substance en solution ou pour suivre l\u2019\u00e9volution des r\u00e9actions dans le temps. Les diff\u00e9rentes cartouches d\u2019\u00e9mission (telles que Emitt.405, Emitt.520, Emitt.635, Emitt.850) permettent \u00e0 l\u2019utilisateur de s\u00e9lectionner la longueur d\u2019onde adapt\u00e9e \u00e0 la substance consid\u00e9r\u00e9e. Ses pics d&rsquo;absorption se situent \u00e0 668 et 609 nm (Merck Index). Diff\u00e9rentes concentrations de bleu de m\u00e9thyl\u00e8ne dans de l&rsquo;eau d\u00e9ionis\u00e9e ont \u00e9t\u00e9 test\u00e9es dans <a href=\"https:\/\/rheolution.com\/products\/absor-b\/\" target=\"_blank\" rel=\"noopener\">l&rsquo;ABSOR.B\u2122<\/a> \u00e0 l&rsquo;aide de flacons en verre transparent de 1 ml et des cartouches Emitt.405, Emitt.520, Emitt.635 et Emitt.850.    <\/p><p><strong>La figure 2<\/strong> pr\u00e9sente les r\u00e9sultats moyens \u00b1 l&rsquo;\u00e9cart-type (trois \u00e9chantillons diff\u00e9rents par concentration test\u00e9e, n = 3). Comme le laissaient pr\u00e9sager les spectres (fl\u00e8ches sur la figure 1), la lumi\u00e8re rouge (635 nm) a \u00e9t\u00e9 fortement absorb\u00e9e, suivie d&rsquo;une l\u00e9g\u00e8re absorption de la lumi\u00e8re verte (520 nm). Les lumi\u00e8res violet-bleu (405 nm) et infrarouge (850 nm) ont pr\u00e9sent\u00e9 une absorption quasi nulle sur toute la plage de concentrations \u00e9tudi\u00e9e.  <\/p><p>La relation entre l&rsquo;absorbance et la concentration est lin\u00e9aire, conform\u00e9ment \u00e0 la <a href=\"https:\/\/chem.libretexts.org\/Bookshelves\/Physical_and_Theoretical_Chemistry_Textbook_Maps\/Supplemental_Modules_(Physical_and_Theoretical_Chemistry)\/Spectroscopy\/Electronic_Spectroscopy\/Electronic_Spectroscopy_Basics\/The_Beer-Lambert_Law\" target=\"_blank\" rel=\"noopener\">loi de Beer-Lambert<\/a>, \u00e0 condition que la solution ne soit pas trop concentr\u00e9e et que la lumi\u00e8re incidente soit monochromatique. La relation lin\u00e9aire obtenue avec la lumi\u00e8re rouge peut \u00eatre utilis\u00e9e pour d\u00e9terminer la concentration en bleu de m\u00e9thyl\u00e8ne dans une solution inconnue. <\/p><p>Conform\u00e9ment aux r\u00e9sultats d&rsquo;absorbance, la transmittance de la lumi\u00e8re rouge (635 nm) diminuait \u00e0 mesure que la concentration en bleu de m\u00e9thyl\u00e8ne dans la solution augmentait. \u00c0 10 mg\/L, moins de 10 % de la lumi\u00e8re \u00e9tait transmise. La transmittance pr\u00e9sentait une l\u00e9g\u00e8re diminution pour les lumi\u00e8res verte (520 nm), bleu-violet (405 nm) et infrarouge (850 nm) (toujours sup\u00e9rieure \u00e0 85 %).  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-305b112 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget
