data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<ul><li>La creciente demanda de m\u00e9todos m\u00e1s r\u00e1pidos para cuantificar c\u00e9lulas va m\u00e1s all\u00e1 de las c\u00e1maras de recuento tradicionales, lo que da lugar a soluciones innovadoras que facilitan un recuento preciso y reducen el tiempo necesario.  <\/li><li>La turbidez, que refleja la opacidad de la soluci\u00f3n debido a la presencia de part\u00edculas que dispersan la luz, resulta ser un m\u00e9todo eficaz para acelerar la cuantificaci\u00f3n celular.<\/li><li>En esta nota de aplicaci\u00f3n, se ha establecido una fuerte correlaci\u00f3n entre la turbidez (FTU) de una soluci\u00f3n de cultivo <em>de S. cerevisiae<\/em> y el n\u00famero de c\u00e9lulas (c\u00e9lulas\/ml) contadas con una c\u00e1mara de recuento celular convencional.  <\/li><li>Usar <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122<\/a> para hacer mediciones precisas de la turbidez te ofrece una forma r\u00e1pida y f\u00e1cil de calcular el n\u00famero de c\u00e9lulas, lo que te elementor-size-default\">Introducci\u00f3n<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-d8b8765 elementor-widget elementor-widget-text-editor\" \u00e1mbito del an\u00e1lisis de cultivos celulares, la necesidad de contar con m\u00e9todos eficaces para cuantificar las c\u00e9lulas ha llevado a buscar alternativas a la t\u00e9cnica tradicional de las c\u00e1maras de recuento celular, que lleva mucho tiempo. Este m\u00e9todo convencional, aunque es fiable, plantea dificultades en cuanto a rapidez y practicidad, sobre todo ante las crecientes exigencias de la investigaci\u00f3n y la industria. <\/p><p>La turbidez, que mide el grado de opacidad de una soluci\u00f3n, se perfila como un enfoque interesante para acelerar la cuantificaci\u00f3n celular. Se ha observado que el nivel de turbidez en un medio est\u00e1 relacionado con la concentraci\u00f3n celular en un cultivo celular [1]. Aprovechando esta relaci\u00f3n, esta nota de aplicaci\u00f3n presenta un nuevo m\u00e9todo para determinar r\u00e1pidamente el n\u00famero de c\u00e9lulas. Este m\u00e9todo aprovecha las capacidades de <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122<\/a>, un instrumento compatible con el IoT dise\u00f1ado para medir la turbidez con precisi\u00f3n. Se inocul\u00f3 un volumen de 100 ml para conseguir un valor inicial de turbidez de 200 FTU. Tras la inoculaci\u00f3n, se tomaron al\u00edcuotas est\u00e9riles cada dos horas y se determin\u00f3 el n\u00famero de c\u00e9lulas (es decir, la concentraci\u00f3n celular) mediante el m\u00e9todo de tinci\u00f3n con azul de metileno. Este m\u00e9todo se basa en un colorante, el azul de metileno, que se adhiere a los componentes celulares y los ti\u00f1e de azul. Este colorante reacciona de forma diferente en las c\u00e9lulas viables y en las no viables, ya que las c\u00e9lulas vivas lo transforman (por lo que permanece incoloro), mientras que las c\u00e9lulas muertas siguen te\u00f1idas de azul. Las c\u00e9lulas te\u00f1idas se colocaron en un <a href=\"https:\/\/en.wikipedia.org\/wiki\/Hemocytometer\" target=\"_blank\" rel=\"noopener\">hemocit\u00f3metro<\/a> (tambi\u00e9n conocido como c\u00e1mara de recuento celular o c\u00e1mara de Malassez) y se observaron bajo el microscopio (x40).      El hemocit\u00f3metro tiene nueve cuadrados de 1 x 1 mm (1 mm\u00b2). Para cada medici\u00f3n se utiliz\u00f3 un volumen de 1 \u00b5L.   <\/p><p>La turbidez de las soluciones se determin\u00f3 con el <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122.<\/a> El instrumento se calibr\u00f3 con patrones de formazina (1-4000 FTU). La emisi\u00f3n y recepci\u00f3n de luz se realizaron utilizando <a href=\"https:\/\/rheolution-store.com\/collections\/cartridges\/products\/emitt-850\" target=\"_blank\" rel=\"noopener\">cartuchos Emitt.805<\/a> (850 nm) y <a href=\"https:\/\/rheolution-store.com\/collections\/cartridges\/products\/cartridge-receiv-vis\" target=\"_blank\" rel=\"noopener\">cartuchos Receiv.ViS<\/a> (longitud de onda entre 400 nm y 1000 nm), respectivamente.   Se utiliz\u00f3 una longitud de onda de 850 nm, ya que la absorci\u00f3n de luz por parte de la muestra era menor (datos no mostrados). En este trabajo, decidimos usar viales de 10 ml, en los que se dividieron las muestras en al\u00edcuotas y se midieron con el <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122.<\/a>  Los datos se recopilaron y se mostraron en la tableta en la que se ejecutaba la aplicaci\u00f3n Soft Matter Analytics App\u2122.<\/p><p>Tanto el recuento celular como las mediciones de turbidez se realizaron por triplicado (n = 3). Los datos se presentan como la media \u00b1 la desviaci\u00f3n est\u00e1ndar. La variabilidad se define como la relaci\u00f3n entre la desviaci\u00f3n est\u00e1ndar y el valor medio.  <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-270ac45 elementor-widget__width-initial elementor-widget-mobile__width-inherit elementor-widget elementor-widget-heading\" data-id=\"270ac45\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">AN\u00c1LISIS COMPARATIVO Y CORRELACI\u00d3N DE LOS PAR\u00c1METROS DE TURBIDEZ Y RECUENTO CELULAR<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-88497d3 elementor-widget elementor-widget-heading\" data-id=\"88497d3\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<p class=\"elementor-heading-title elementor-size-default\">Resultados y discusi\u00f3n<\/p>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-a479882 elementor-widget elementor-widget-text-editor\" data-id=\"a479882\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Se analizaron al\u00edcuotas celulares en la fase de crecimiento del cultivo de levadura midiendo la turbidez y contando las c\u00e9lulas con una c\u00e1mara de recuento celular. <b>La figura 3<\/b> muestra la evoluci\u00f3n temporal de ambos par\u00e1metros, que aumentan con el tiempo. En el gr\u00e1fico se aprecia que la evoluci\u00f3n de la turbidez y la concentraci\u00f3n celular siguen la misma tendencia, que en general es exponencial. El aumento de la concentraci\u00f3n celular influye directamente en la turbidez del cultivo. Cuantas m\u00e1s c\u00e9lulas hay, m\u00e1s luz se dispersa y mayor es la turbidez general del medio.   <\/p><p>Las mediciones de turbidez fueron m\u00e1s consistentes (con una variabilidad del 3,8 %) en comparaci\u00f3n con la t\u00e9cnica de recuento celular (con una variabilidad del 17,6 %). Esta diferencia se puede atribuir a imprecisiones en el recuento debidas a la percepci\u00f3n humana, pero tambi\u00e9n a la diferencia en el volumen de muestra entre los dos m\u00e9todos. Mientras que el recuento celular utiliza 1 \u00b5l de muestra, en este estudio la evaluaci\u00f3n de la turbidez se realiza con al\u00edcuotas de 10 ml. El gr\u00e1fico obtenido se puede utilizar luego para estimar el recuento celular de una muestra, simplemente correlacionando el valor de turbidez medido con <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">TURBIDI.T\u2122<\/a> con el n\u00famero de c\u00e9lulas correspondiente.  Este m\u00e9todo de inferencia consta de cuatro pasos sucesivos (<b>Figura 5<\/b>):<\/p><ol><li>Se reparten al\u00edcuotas de una muestra que contiene una concentraci\u00f3n desconocida de c\u00e9lulas en uno o varios viales.<\/li><li>Se coloca la probeta en la c\u00e1mara <a href=\"https:\/\/rheolution.com\/life-sciences\/products\/turbidi-t\/\" target=\"_blank\" rel=\"noopener\">del TURBIDI.T\u2122<\/a> y se realiza una medici\u00f3n. Los resultados se muestran en la aplicaci\u00f3n Soft Matter Analytics\u2122. <\/li><li>Al usar la curva de correlaci\u00f3n (c\u00e9lulas\/mL frente a FTU), las FTU se relacionan con la densidad
