Es una magnitud adimensional que se calcula a partir del logaritmo de la relaci\u00f3n entre la luz incidente y la luz transmitida a trav\u00e9s de una muestra. El espectro de absorbancia, que suele representarse en un gr\u00e1fico, muestra las longitudes de onda en las que una sustancia absorbe la luz con mayor intensidad (<strong>Figura 1<\/strong>). Esta informaci\u00f3n resulta \u00fatil en diversos campos cient\u00edficos, como la biolog\u00eda, la qu\u00edmica, la bioqu\u00edmica y la biolog\u00eda molecular, para identificar y cuantificar compuestos o mol\u00e9culas espec\u00edficos en una muestra.   <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-e987a6c Los valores de absorbancia en una longitud de onda concreta se usan mucho en an\u00e1lisis qu\u00edmicos y bioqu\u00edmicos para cuantificar la concentraci\u00f3n de una sustancia en soluci\u00f3n o para seguir la evoluci\u00f3n de las reacciones a lo largo del tiempo. Los diferentes cartuchos de emisi\u00f3n (como el Emitt.405, el Emitt.520, el Emitt.635 y el Emitt.850) te permiten elegir la longitud de onda adecuada para cada sustancia. Sus picos de absorci\u00f3n est\u00e1n a 668 y 609 nm (Merck Index). Se probaron diferentes concentraciones de azul de metileno en agua desionizada en el <a href=\"https:\/\/rheolution.com\/products\/absor-b\/\" target=\"_blank\" rel=\"noopener\">ABSOR.B\u2122<\/a> utilizando viales de cristal transparente de 1 ml y los cartuchos Emitt.405, Emitt.520, Emitt.635 y Emitt.850.    <\/p><p><strong>La figura 2<\/strong> muestra los resultados medios \u00b1 desviaci\u00f3n est\u00e1ndar (tres muestras diferentes por cada concentraci\u00f3n analizada, n = 3). Tal y como cab\u00eda esperar a partir del espectro (flechas de la figura 1), la luz roja (635 nm) se absorbi\u00f3 en gran medida, seguida de una ligera absorci\u00f3n de la luz verde (520 nm). Las luces violeta-azul (405 nm) e infrarroja (850 nm) tuvieron una absorci\u00f3n casi nula en todo el rango de concentraciones estudiado.  <\/p><p>La relaci\u00f3n entre la absorbancia y la concentraci\u00f3n es lineal seg\u00fan la <a href=\"https:\/\/chem.libretexts.org\/Bookshelves\/Physical_and_Theoretical_Chemistry_Textbook_Maps\/Supplemental_Modules_(Physical_and_Theoretical_Chemistry)\/Spectroscopy\/Electronic_Spectroscopy\/Electronic_Spectroscopy_Basics\/The_Beer-Lambert_Law\" target=\"_blank\" rel=\"noopener\">ley de Beer-Lambert<\/a>, siempre que la soluci\u00f3n no sea demasiado concentrada y la luz incidente sea monocrom\u00e1tica. La relaci\u00f3n lineal que se obtiene con la luz roja se puede usar para calcular la concentraci\u00f3n de azul de metileno en una soluci\u00f3n desconocida. <\/p><p>Seg\u00fan los resultados de absorbancia, la transmitancia de la luz roja (635 nm) disminuy\u00f3 a medida que aumentaba la concentraci\u00f3n de azul de metileno en la soluci\u00f3n. A 10 mg\/L, se transmit\u00eda menos del 10 % de la luz. La transmitancia disminuy\u00f3 ligeramente para las luces verde (520 nm), azul violeta (405 nm) e infrarroja (850 nm) (siempre por encima del 85 %).
